VOB560 + MIK665 is a combination of two experimental small molecule drugs developed to treat hematological malignancies by simultaneously inhibiting anti-apoptotic proteins in cancer cells. VOB560 is a selective B-cell lymphoma 2 (BCL2) inhibitor, while MIK665 is a selective myeloid cell leukemia 1 (MCL1) inhibitor. These agents act as BH3 mimetics that block the respective proteins, leading to apoptosis of tumor cells that otherwise escape cell death. In preclinical and early clinical studies, the combination has shown synergistic anti-tumor activity, specifically in patients with relapsed or refractory non-Hodgkin lymphoma, acute myeloid leukemia, and multiple myeloma. The drugs are administered intravenously and were investigated in a phase 1b, multi-center trial to establish safety, tolerability, and preliminary efficacy[1][3][5].
