Overview

**vobramitamab duocarmazine + MGA012** is an experimental combination of two investigational monoclonal antibody-based oncology agents developed by MacroGenics. **Vobramitamab duocarmazine** (MGC018, also called vobra duo) is an antibody-drug conjugate (ADC) targeting B7-H3, a protein commonly overexpressed in solid tumors. It delivers a **duocarmycin**-based cytotoxic payload via a cleavable peptide linker to tumor cells, causing DNA alkylation and cell death. **MGA012** (also known as INCMGA00012) is a monoclonal antibody that blocks PD-1, a key immune checkpoint receptor, thereby antagonizing PD-1/PD-L1 interaction and enabling T cell-mediated anti-tumor immune responses. The combination is evaluated as an anti-cancer therapy in advanced solid tumors, aiming for a dual approach: direct cytotoxicity and immune checkpoint blockade. As of 2024, treatment development has been discontinued due to insufficient efficacy and concerns about toxicity for vobramitamab duocarmazine[1][3][4][5].
