Drug intelligence / Profile preview
MGC026 + pembrolizumab is an investigational combination regimen pairing vobramitamab tavatecan (MGC026), a B7-H3-directed antibody-drug conjugate (ADC), with pembrolizumab, an anti-PD-1 monoclonal antibody. Vobramitamab tavatecan consists of a humanized anti-B7-H3 monoclonal antibody (MGA017) site-specifically conjugated to a cleavable exatecan-based topoisomerase I (TOP1) inhibitor payload (SYNtecan E) with a drug-to-antibody ratio (DAR) of 4. Pembrolizumab is a well-established immune checkpoint inhibitor that blocks the interaction between PD-1 and its ligands, PD-L1 and PD-L2, thereby revitalizing T-cell-mediated antitumor immune responses. The combination is being developed by MacroGenics for the treatment of advanced solid tumors, with plans to initiate a Phase 1 expansion cohort in early 2027 for front-line patients with PD-L1-positive recurrent or metastatic squamous cell carcinoma of the head and neck (SCCHN).
