Drug intelligence / Profile preview
Voclosporin ophthalmic solution (VOS) is an investigational, aqueous, preservative-free nanomicellar formulation of voclosporin, a potent calcineurin inhibitor (CNI) developed by Aurinia Pharmaceuticals for the treatment of dry eye syndrome. As a small molecule immunosuppressant, voclosporin functions by blocking the expression of interleukin-2 (IL-2) and inhibiting T-cell mediated immune responses that contribute to ocular surface inflammation. Although Phase 2 trials initially suggested VOS might offer superior efficacy compared to standard treatments like Restasis, the pivotal Phase 2/3 AUDREY trial failed to meet its primary endpoint of a ≥10mm improvement in the Schirmer Tear Test at 4 weeks. Following these results, Aurinia Pharmaceuticals discontinued the clinical development program for VOS in dry eye syndrome in November 2020.
