**Vofatamab + pembrolizumab** is an investigational combination therapy developed for the treatment of locally advanced or metastatic urothelial cell carcinoma (bladder cancer) in patients who have progressed following platinum-based chemotherapy and have not received prior immune checkpoint inhibitors[1][3][5][7]. Vofatamab is a fully human monoclonal antibody targeting fibroblast growth factor receptor 3 (FGFR3), blocking activation of both wild-type and genetically altered (mutant or fusion) FGFR3, thereby inhibiting FGFR3-driven tumor proliferation and survival[1][2][5]. Pembrolizumab is an immune checkpoint inhibitor that targets programmed cell death protein 1 (PD-1), reactivating T-cell-mediated immune responses against tumor cells. This combination aims to extend clinical benefit to both FGFR3-mutated and wild-type patients, including those less likely to respond to immune checkpoint inhibition alone, and has shown an overall response rate (ORR) of about 40% in early-phase clinical trials[5].
