vonoprazan + clarithromycin is a dual therapy combination regimen being investigated by Peking University First Hospital for the first-line eradication of Helicobacter pylori infection. This simplified regimen consists of vonoprazan, a potassium-competitive acid blocker (P-CAB), and clarithromycin, a macrolide antibiotic. Vonoprazan provides rapid and sustained gastric acid suppression by competitively inhibiting the H+, K+-ATPase pump, which enhances the stability and antimicrobial activity of clarithromycin. Clarithromycin exerts its effect by binding to the 50S ribosomal subunit of the bacteria, thereby inhibiting protein synthesis. This specific study evaluates the efficacy of the dual therapy in patients pre-screened for a lack of clarithromycin resistance using fecal gene detection.
