Vonoprazan + itopride is an investigational combination therapy being evaluated for the treatment of functional dyspepsia (FD). Vonoprazan is a potassium-competitive acid blocker (PCAB) that provides potent and sustained suppression of gastric acid by inhibiting the H+/K+-ATPase pump. Itopride is a prokinetic agent with a dual mechanism of action involving dopamine D2 receptor antagonism and acetylcholinesterase inhibition, which enhances gastrointestinal motility. The combination is designed to address both acid-related and motility-related symptoms of FD, such as epigastric pain, postprandial fullness, and early satiety. It is currently being studied in the VIVA Trial (Vonoprazan vs Vonoprazan + Itopride in Functional Dyspepsia) sponsored by the Asian Institute of Gastroenterology in India.
