Vopimetostat + ERAS-0015 is an investigational combination therapy comprising vopimetostat (TNG462), a next-generation PRMT5 inhibitor, and ERAS-0015, a pan-RAS molecular glue. Vopimetostat is designed to selectively target MTAP-deleted cancer cells by exploiting the accumulation of methylthioadenosine (MTA), which sensitizes these cells to PRMT5 inhibition. ERAS-0015 acts by recruiting cyclophilin A to RAS proteins, thereby inhibiting signaling across multiple RAS mutations and disrupting the MAPK/ERK and PI3K/AKT pathways. The combination is being developed under a clinical trial collaboration between Tango Therapeutics and Erasca to treat MTAP-deleted pancreatic cancer and RAS-mutant non-small cell lung cancer (NSCLC), aiming to provide synergistic anti-tumor activity by simultaneously blocking the RAS pathway and PRMT5.
