VOR33 + gemtuzumab ozogamicin is a combination therapeutic approach under investigation for the treatment of acute myeloid leukemia (AML). **VOR33** is an allogeneic, CRISPR/Cas9 genome-edited hematopoietic stem and progenitor cell (HSPC) therapy engineered to lack the **CD33** protein, making patient hematopoietic systems resistant to CD33-directed therapies post-transplant. **Gemtuzumab ozogamicin** is an antibody-drug conjugate (ADC) consisting of a monoclonal antibody targeting CD33 conjugated to the cytotoxic agent calicheamicin. This approach enables selective targeting and killing of CD33-positive leukemic cells with gemtuzumab ozogamicin after transplantation with VOR33-engineered HSPCs, sparing healthy engrafted cells that lack CD33[1][5][2][9][10].
