Overview

Voriconazole + amphotericin B is an antifungal combination therapy used primarily for the treatment of invasive fungal infections, particularly refractory aspergillosis. This combination leverages the distinct mechanisms of action of both drugs: voriconazole alters cell membrane function by inhibiting ergosterol synthesis, while amphotericin B binds to ergosterol in the fungal cell membrane, creating transmembrane channels that disrupt membrane integrity. The combination is sometimes employed as primary or salvage therapy with the goal of improving monotherapy outcomes, especially in immunocompromised patients with severe or resistant fungal infections.
