Overview

Vorinostat + carboplatin + gemcitabine is a combination chemotherapy and epigenetic therapy regimen studied for the treatment of recurrent, platinum-sensitive epithelial ovarian, fallopian tube, and peritoneal cancers. Vorinostat (Zolinza) is a small molecule histone deacetylase (HDAC) inhibitor that stops cancer cell growth by interacting with chromosomes and may sensitize cells to platinum-based agents. Carboplatin is a platinum-based DNA alkylating agent, and gemcitabine is a nucleoside analog that inhibits DNA synthesis. This specific combination was evaluated in a Phase Ib/II trial led by the Dana-Farber Cancer Institute to determine the maximally tolerated dose and efficacy. However, the trial was terminated early due to significant hematologic toxicities, including grade 3-4 neutropenia and thrombocytopenia, and no maximally tolerated dose was established.
