A four-drug combination regimen consisting of vorinostat (a histone deacetylase inhibitor), lenalidomide (an immunomodulatory agent), carfilzomib (a second-generation proteasome inhibitor), and dexamethasone (a corticosteroid). This combination leverages the synergistic anti-multiple myeloma activity of these agents with different mechanisms of action. The regimen has been studied in phase I clinical trials for relapsed and/or refractory multiple myeloma.
