vorinostat + temsirolimus is a combination therapy investigated by the Roswell Park Cancer Institute for the treatment of metastatic hormone-refractory, chemoresistant prostate cancer. Vorinostat (also known as SAHA or Zolinza) is a small molecule histone deacetylase (HDAC) inhibitor that works by inhibiting enzymes responsible for the deacetylation of lysine residues on histones and non-histone proteins, thereby regulating gene expression and inducing cell cycle arrest or apoptosis. Temsirolimus (also known as CCI-779 or Torisel) is a small molecule inhibitor of the mammalian target of rapamycin (mTOR), a kinase that plays a central role in cell growth, proliferation, and survival. The combination of these two agents is designed to target multiple signaling pathways and epigenetic mechanisms to overcome resistance in advanced prostate cancer. A Phase I clinical trial (NCT01174199) was conducted to evaluate the safety, tolerability, and optimal dosing of this combination in patients who had progressed after docetaxel treatment.
