Voyager-V1 + pembrolizumab + ipilimumab + nivolumab is an investigational combination immunotherapy regimen developed by Vyriad for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC), neuroendocrine carcinoma (NEC), and head and neck cancer. The regimen combines Voyager-V1 (VV1)—an oncolytic vesicular stomatitis virus (VSV) engineered to express human interferon-beta (IFNβ) to enhance anti-tumor immune responses and the human sodium iodide symporter (NIS) for non-invasive SPECT/PET imaging—with immune checkpoint inhibitors. These checkpoint inhibitors include pembrolizumab (an anti-PD-1 monoclonal antibody) or the combination of ipilimumab (an anti-CTLA-4 monoclonal antibody) and nivolumab (an anti-PD-1 monoclonal antibody). By combining direct viral-mediated oncolysis and localized interferon-beta expression with systemic immune checkpoint blockade, the regimen aims to convert immunologically 'cold' tumors into 'hot' tumors, thereby enhancing anti-tumor T-cell responses and overcoming resistance to single-agent checkpoint inhibition.
