Overview

VRC-HIVDNA016-00-VP is an investigational DNA plasmid vaccine developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID) for the prevention of HIV-1 infection. The vaccine consists of a mixture of six mammalian expression plasmids (using the pVRC8400 backbone) that encode HIV-1 Gag, Pol, and Nef proteins from Clade B, as well as Env proteins from Clades A, B, and C. When injected intramuscularly, the plasmids enter host cells where the encoded viral antigens are expressed and presented to the immune system, primarily to elicit cell-mediated immune responses (CD8+ T-cells) and prime the immune system for subsequent booster vaccinations. It has been evaluated in Phase 1 clinical trials, including studies examining host genetics in twin pairs, often as part of a heterologous prime-boost regimen with recombinant adenoviral vectors.
