VRC01 + combination antiretroviral therapy (cART) is an investigational treatment regimen for HIV-1 infection, specifically studied in pediatric populations by the National Institute of Allergy and Infectious Diseases (NIAID). VRC01 is a human broadly neutralizing monoclonal antibody (bNAb) that targets the highly conserved CD4-binding site of the HIV-1 envelope glycoprotein gp120, thereby neutralizing a wide range of HIV-1 strains and potentially facilitating the clearance of infected cells. When used in conjunction with cART—which typically consists of a cocktail of antiretroviral drugs such as reverse transcriptase inhibitors, protease inhibitors, or integrase inhibitors—the regimen aims to achieve deeper viral suppression and reduce the size of the latent viral reservoir. This combination is being evaluated for its safety, pharmacokinetics, and ability to promote immune-mediated clearance of HIV-1 in infants starting therapy shortly after birth.
