VRC07-523LS + vorinostat is a combination therapy investigated by the University of North Carolina at Chapel Hill as part of a "shock and kill" strategy for HIV-1 cure. The regimen consists of vorinostat (Zolinza), a small molecule histone deacetylase (HDAC) inhibitor, and VRC07-523LS, a potent broadly neutralizing monoclonal antibody (bNAb). Vorinostat acts as a latency-reversing agent (LRA) to induce the expression of latent HIV-1 proviruses from resting CD4+ T cells ("shock"). VRC07-523LS targets the CD4 binding site of the HIV-1 envelope glycoprotein gp120 and is intended to neutralize the reactivated virus and facilitate the clearance of infected cells through immune-mediated mechanisms ("kill"). Clinical studies have evaluated the safety and efficacy of this combination in HIV-positive individuals on suppressive antiretroviral therapy.
