VRP-HER2 + pembrolizumab is an investigational combination therapy for HER2-positive cancers, particularly breast cancer. VRP-HER2 is a vaccine based on a Venezuelan equine encephalitis virus replicon particle (VRP) platform encoding the HER2 antigen, designed to stimulate an immune response against HER2-expressing tumor cells. Pembrolizumab is a monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor and acts as an immune checkpoint inhibitor to enhance T-cell mediated anti-tumor activity. The combination aims to prime and boost anti-tumor immunity by both generating HER2-specific T-cell responses via vaccination and relieving immunosuppression through PD-1 blockade. This approach is being evaluated in phase II clinical trials for patients with advanced or metastatic HER2-positive breast cancer[4][5][6][7].
