VS-411 is a small molecule vitamin D receptor modulator (VDRM) developed by Vidasym, a clinical-stage biotechnology company. It functions as a potent agonist of the vitamin D receptor (VDR), demonstrating high affinity and activity in reporter gene assays with an EC50 of less than 0.1 nM. Preclinical research in uremic rat models showed that VS-411 effectively suppresses parathyroid hormone (PTH) levels, indicating potential for treating secondary hyperparathyroidism in patients with chronic kidney disease (CKD). However, the compound exhibited a narrow therapeutic index in animal studies due to hypercalcemic effects. Although it has been considered for other inflammatory conditions such as psoriasis and inflammatory bowel disease, its current development status remains unclear, as it is listed without specific indications or clinical phases in recent company pipelines.
