This is a **combination therapy** consisting of *VT3989*, a small molecule TEAD transcriptional enhanced associate domain (TEAD) autopalmitoylation inhibitor, plus *nivolumab* (a PD-1 checkpoint inhibitor monoclonal antibody) and *ipilimumab* (a CTLA-4 checkpoint inhibitor monoclonal antibody). VT3989 targets the Hippo pathway by inhibiting TEAD proteins, as well as Focal adhesion kinase (FAK) and Proline-rich tyrosine kinase 2 (PYK2), disrupting survival pathways in cancer cells. Nivolumab and ipilimumab are immune checkpoint inhibitors that enhance T-cell mediated anti-tumor responses by blocking PD-1 and CTLA-4, respectively. This combination is being investigated primarily for mesothelioma, especially in cases with recurrence or progression after standard treatment, and may have additional potential for other solid tumors[1][3][5].
