Vusolimogene oderparepvec + nivolumab is an **investigational combination therapy** for advanced melanoma that pairs a replication-selective, genetically engineered oncolytic virus (**vusolimogene oderparepvec, also known as RP1**) with a checkpoint inhibitor (**nivolumab**, an anti–PD-1 antibody)[1][2][4][5][8]. Vusolimogene oderparepvec is a modified herpes simplex virus type 1 that selectively infects and lyses tumor cells, induces immunogenic tumor cell death, and stimulates systemic anti-tumor immunity. It is armed with additional immunostimulatory payloads (fusogenic GALV-CPR and GM-CSF) intended to further enhance immune activation and tumor destruction. Nivolumab is a monoclonal antibody that targets and blocks the programmed death 1 (**PD-1**) protein on T cells, reactivating anti-tumor immune responses. The combination is delivered via intratumoral (vusolimogene oderparepvec) and intravenous (nivolumab) administration.
