Drug intelligence / Profile preview
VVD-214 + bevacizumab is an investigational combination therapy being evaluated for the treatment of advanced microsatellite instability-high (MSI-high) or deficient mismatch repair (dMMR) colorectal cancer. VVD-214 (also known as RO7589831 or VVD-133214) is an oral, small-molecule, covalent allosteric inhibitor of Werner helicase (WRN), a DNA repair enzyme that represents a synthetic lethal target in MSI-high or dMMR cancers. Bevacizumab (Avastin) is a recombinant humanized monoclonal antibody that targets vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and tumor vascularization. The combination is being evaluated in a Phase 1b clinical trial (NCT06004245) in patients with previously treated advanced MSI-high or dMMR colorectal cancer.
