Overview

WGI-0301 + sorafenib is an investigational combination therapy for advanced hepatocellular carcinoma (HCC). **WGI-0301** is a lipid nanoparticle formulation of Archexin, a 20-mer phosphorothioate antisense oligonucleotide targeting Akt-1 mRNA, thereby inhibiting the translation and activity of Akt-1, a serine/threonine kinase involved in cell proliferation, survival, angiogenesis, and resistance pathways. WGI-0301 is designed for targeted delivery using the QTsome nucleic acid delivery technology, enhancing intracellular uptake and specificity for tumor cells. **Sorafenib** is an oral multi-kinase inhibitor that targets tumor cell proliferation and angiogenesis by inhibiting several kinases, including RAF kinases and vascular endothelial growth factor receptors (VEGFR). The combination is under phase 2 clinical investigation for treatment of advanced, unresectable HCC, especially in patients previously treated with immune checkpoint inhibitors. Preclinical and early clinical studies demonstrate that combining WGI-0301 with sorafenib can augment anti-tumor and anti-angiogenic efficacy compared to sorafenib alone, without significantly increasing toxicity[1][2][3][5][7][9].
