This is an autologous dendritic cell (DC) vaccine developed by Antwerp University Hospital (UZA). The vaccine consists of patient-derived dendritic cells that have been ex vivo electroporated with messenger RNA (mRNA) encoding the Wilms tumor 1 (WT1) protein, along with mRNA for interleukin-15 (IL-15) and the interleukin-15 receptor alpha (IL-15Rα). WT1 serves as a widely expressed tumor-associated antigen, while the co-expression of IL-15 and IL-15Rα is designed to facilitate the trans-presentation of IL-15 to neighboring immune cells. This mechanism aims to enhance the activation, survival, and proliferation of cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells within the tumor microenvironment, thereby mounting a more robust anti-tumor immune response. It is primarily being investigated for advanced solid tumors, including pancreatic, esophageal, liver, and ovarian cancers.
