WOKVAC + pertuzumab + trastuzumab is an investigational combination therapy being studied primarily for the treatment of HER2-positive breast cancer. This regimen combines three agents: - **WOKVAC** is a therapeutic cancer vaccine composed of tumor-associated antigens, designed to stimulate the immune system to recognize and attack HER2-expressing tumor cells. - **Pertuzumab** is a humanized monoclonal antibody that binds to subdomain II of the HER2 receptor, inhibiting its dimerization with other HER family members and blocking downstream signaling pathways involved in cell growth and survival. - **Trastuzumab** is another humanized monoclonal antibody targeting subdomain IV of the HER2 receptor, preventing activation and promoting immune-mediated destruction (antibody-dependent cellular cytotoxicity) of tumor cells. The combination aims to enhance anti-tumor efficacy by simultaneously stimulating antigen-specific T-cell responses (via WOKVAC) and providing dual blockade of the HER2 pathway (via pertuzumab and trastuzumab). This approach may improve pathological response rates when given as neoadjuvant therapy before surgery in patients with early-stage or locally advanced HER2-positive breast cancer[1][4][8].
