WP1122 + WP1096 + WP1097 is a preclinical-stage small molecule combination portfolio developed by Moleculin Biotech. These three compounds collectively form the 'WP1122 Portfolio' of D-glucose and D-mannose antimetabolites, designed to exploit the potential uses of monosaccharide decoys (such as glucose or mannose) as a means to inhibit glycolysis and alter glycosylation — effectively cutting off the energy supply of tumors and viruses. WP1122 is a prodrug of 2-deoxy-D-glucose (2-DG), which is designed to improve the drug-like properties of 2-DG by increasing its circulation time and improving tissue/organ distribution. WP1096 and WP1097 are structurally distinct analogs within this portfolio that have demonstrated significant in vitro antiviral activity against SARS-CoV-2, HIV, Zika, and Dengue Fever. WP1096 has also been selected for NIAID-funded animal studies targeting hemorrhagic fever arenaviruses (HFA). The portfolio is being developed for the potential treatment of viral infections (including COVID-19, HIV, Zika, Dengue, and hemorrhagic fevers) and various cancers.
