This investigational therapy is an autologous dendritic cell-based vaccine designed to stimulate a multi-pronged immune response against three highly conserved tumor-associated antigens: Wilms' Tumor 1 (WT1), Human Telomerase Reverse Transcriptase (hTERT), and Survivin. The vaccine is produced by harvesting a patient's peripheral blood mononuclear cells, differentiating them into dendritic cells, and pulsing or transfecting them with peptides or mRNA corresponding to the three target antigens. Once re-administered to the patient, these mature dendritic cells migrate to the lymph nodes to present the antigens to naive T-lymphocytes, thereby activating and expanding antigen-specific CD8+ cytotoxic T-cells and CD4+ helper T-cells. This multi-antigen approach is intended to enhance the breadth and durability of the anti-tumor immune response, addressing the challenge of tumor heterogeneity and potential antigen escape common in aggressive malignancies such as glioblastoma and acute myeloid leukemia.
