Overview

This investigational peptide vaccine was developed by Radboud University Nijmegen Medical Centre for the treatment of acute myeloid leukemia (AML) and multiple myeloma (MM). The vaccine is composed of HLA-A*0201-restricted peptides derived from three tumor-associated antigens: Wilms Tumor protein (WT1), Proteinase 3 (PR3), and Mucin 1 (MUC1). It also includes the pan HLA-DR T helper cell epitope (PADRE) to provide CD4+ T cell help. To enhance the immune response, the peptides were formulated with the adjuvants Montanide ISA51 and the TLR9 agonist CpG7909. The vaccine was designed to induce tumor-specific CD8+ T cell responses capable of lysing cancer cells. However, clinical trial results were disappointing, showing no clinical responses and a significant decline in vaccine-specific CD8+ T cells, leading to the conclusion that this specific combination of peptides and adjuvants might be detrimental to the immune response.
