WT1-A10 + AS01B is an investigational therapeutic cancer vaccine developed by GSK, consisting of a recombinant Wilms Tumor protein 1 (WT1) fusion protein formulated with the proprietary AS01B adjuvant system. The antigen, WT1-A10, is a truncated recombinant protein covering amino acids 2 to 441 of the human WT1 transcription factor, which is highly overexpressed in various hematological malignancies and solid tumors but has limited expression in normal tissues. The AS01B adjuvant is a liposomal formulation containing two key immunostimulants: 3-O-desacyl-4'-monophosphoryl lipid A (MPL), which acts as a TLR4 agonist, and the saponin QS-21. This combination is designed to enhance the magnitude and duration of the immune response, specifically by inducing antigen-specific CD4+ and CD8+ T-cell responses against cells expressing WT1. Although it reached Phase 2 clinical trials for conditions like acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS), development was eventually terminated by GSK as part of a strategic shift in their oncology pipeline.
