**WTX-124 + mWTX-330** is a combination of two conditionally activated INDUKINE™ cytokine therapies from Werewolf Therapeutics designed for cancer immunotherapy. **WTX-124** is a systemically delivered prodrug of recombinant interleukin-2 (IL-2) that remains inactive in peripheral tissues but activates selectively in the tumor microenvironment (TME) via tumor protease-sensitive linkers, locally stimulating T-cell proliferation and antitumor immune responses while minimizing systemic toxicities. **mWTX-330** is a mouse surrogate for WTX-330, comprising chimeric IL-12 (mouse p35 and human p40 subunits) tethered to a half-life extension domain and inactivation domain by protease-cleavable linkers, enabling selective intratumoral activation to boost cross-presenting dendritic cells, natural killer cells, CD8+ T cells, and reshape the TME toward a TH1 phenotype without peripheral toxicity. The combination leverages synergistic cytokine effects for enhanced efficacy in advanced solid tumors, with preclinical sequential administration noted.[2][3][7]
