XB010 (Exelixis, Inc.)

XB010 (also known as DLL3-CSL20-Exa) is an investigational antibody-drug conjugate (ADC) developed by Exelixis. It is designed to target Delta-like ligand 3 (DLL3), an inhibitory Notch ligand that is highly expressed on the surface of small cell lung cancer (SCLC) and other neuroendocrine carcinomas, while showing minimal expression in normal tissues. The ADC consists of a humanized monoclonal antibody (clone CSL20) covalently linked to a potent exatecan-derivative payload, which acts as a DNA topoisomerase I inhibitor. The linker is designed to be stable in systemic circulation but susceptible to cleavage by lysosomal proteases within the tumor cell. Upon binding to DLL3 and subsequent internalization, the payload is released, inducing DNA damage and apoptosis in the target cell and potentially exerting a bystander effect on neighboring tumor cells. XB010 is currently being evaluated in Phase 1 clinical trials for patients with advanced solid tumors associated with DLL3 expression.
