Overview
This is a **combination regimen** consisting of XB2001 (an investigational monoclonal antibody targeting Connective Tissue Growth Factor, CTGF), liposomal irinotecan (Onivyde, a liposomal formulation of irinotecan—a topoisomerase I inhibitor), 5-fluorouracil (a thymidylate synthase inhibitor), and leucovorin (folinic acid, which stabilizes the bond of 5-FU with TS). Liposomal irinotecan with 5-fluorouracil and leucovorin is an approved regimen for metastatic pancreatic cancer after progression on gemcitabine-based therapy[1][3][4]. XB2001 is being studied in combination with this backbone for its potential to target tumor stroma and improve chemotherapy delivery. The mechanism of the regimen combines cytotoxic activity against tumor DNA/RNA synthesis (5-FU, irinotecan), modulation of chemotherapy activity (leucovorin), and possible anti-stroma, anti-CTGF effects (XB2001 if included). Indication: being studied primarily for pancreatic adenocarcinoma.
