Xenopax + glucocorticoid is a combination therapy being evaluated as a first-line treatment for acute graft-versus-host disease (aGVHD) following allogeneic hematopoietic stem cell transplantation (allo-HSCT). The combination consists of Xenopax, a humanized monoclonal antibody that targets the alpha subunit of the interleukin-2 receptor (CD25) expressed on activated T lymphocytes, and glucocorticoids, which provide broad anti-inflammatory and immunosuppressive effects. By blocking the IL-2 receptor, Xenopax inhibits the proliferation of T-cells involved in the pathogenesis of aGVHD. This therapeutic approach is being investigated by the Institute of Hematology & Blood Diseases Hospital in China to improve clinical outcomes and remission rates in patients with aGVHD compared to standard steroid monotherapy. Xenopax has the same amino acid sequence as daclizumab and is approved in China for the prevention of acute rejection after kidney transplantation.
