Drug intelligence / Profile preview
Neotri is an oral fixed-dose combination of the thiazide-like diuretic xipamide and the potassium-sparing diuretic triamterene. Originally developed by Eli Lilly and currently owned and manufactured by Mibe Arzneimittel (a subsidiary of Dermapharm), Neotri is approved in Germany for the treatment of mild-to-moderate hypertension and various forms of edema (fluid retention) associated with cardiac, hepatic, renal, or venous disorders. Xipamide acts primarily on the distal convoluted tubule by inhibiting the sodium-chloride cotransporter (NCC/SLC12A3), promoting the excretion of sodium and water. Triamterene acts on the late distal tubule and collecting duct by blocking epithelial sodium channels (ENaC), which reduces sodium reabsorption and limits the excretion of potassium, thereby counteracting the hypokalemic effects typically associated with thiazide-like diuretics.
