Overview

**XNW5004 + pembrolizumab** is a combination investigational therapy developed for the treatment of advanced solid tumors and other cancers. XNW5004 is a rationally designed, potent, and selective small molecule inhibitor of the enzyme **enhancer of zeste homolog 2 (EZH2)**, an epigenetic regulator implicated in tumorigenesis, tumor progression, and resistance to immunotherapy. Pembrolizumab (KEYTRUDA) is a well-established immunotherapy and humanized monoclonal antibody that targets the **programmed cell death protein 1 (PD-1)** receptor, enhancing the anti-tumor immune response. Preclinical and early clinical data indicate that XNW5004 may synergize with anti-PD-1 therapy to improve immune-mediated tumor control. The combination is currently under investigation in Phase 1/2 trials for patients with advanced solid tumors who have failed standard treatments, with Evopoint Biosciences sponsoring and Merck supplying pembrolizumab[1][2][3][5].
