XP-DC + tumor lysate + keyhole limpet hemocyanin is an autologous dendritic cell-based immunotherapy developed by Radboud University Medical Center (Radboudumc) for the treatment of ovarian cancer. The therapy involves isolating monocytes from a patient's blood and differentiating them ex vivo into dendritic cells (DCs). These cells are matured using a specific cytokine cocktail (often referred to as the 'XP' cocktail, containing TNF-α, IL-1β, IL-6, and PGE2) in X-VIVO 15 medium. The resulting mature DCs (XP-DCs) are then loaded (pulsed) with autologous tumor lysate, which provides a broad spectrum of patient-specific tumor-associated antigens, and keyhole limpet hemocyanin (KLH), which acts as an immunogenic carrier and adjuvant. The vaccine is typically administered via intranodal injection to stimulate a robust, multi-antigenic T-cell response intended to recognize and eliminate ovarian cancer cells.
