Overview

Yellow fever vaccine (17D strain) is a live-attenuated viral vaccine used for active immunization against yellow fever. Originally developed in 1937 by Max Theiler at the Rockefeller Foundation, the vaccine is derived from the wild-type Asibi strain of the yellow fever virus through serial passages in mouse and chicken embryo tissues. This process yielded the attenuated 17D strain, which has since been maintained as three primary substrains: 17D-204, 17DD, and 17D-213. The vaccine is manufactured by culturing these substrains in embryonated chicken eggs. It is highly immunogenic, with a single subcutaneous or intramuscular dose eliciting protective levels of neutralizing antibodies and robust T-cell responses in over 99% of recipients within 30 days, providing lifelong immunity for most individuals. The vaccine is marketed globally under various brand names, including YF-VAX and Stamaril by Sanofi, and is produced by a limited number of WHO-prequalified manufacturers to support global immunization programs and travel prophylaxis.
