YL-13027 + sintilimab is a combination therapy being investigated for advanced solid tumors. YL-13027 is a novel small molecule inhibitor targeting the type 1 receptor of transforming growth factor beta (TGF-βR1), also known as ALK5, and is designed to neutralize TGF-β signaling in the tumor microenvironment, thereby modulating immunosuppression and potentially enhancing antitumor activity. Sintilimab is a fully human IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor. The combination is theorized to synergistically promote immune activity against tumors by both reducing immunosuppressive TGF-β signaling and blocking PD-1 mediated T cell inhibition. This combination is currently being evaluated in a multicenter, open-label phase 1b/2 clinical trial in patients with advanced solid tumors to determine safety, optimal dosing, and preliminary efficacy[1][2][3][5][7].
