YL201 + atezolizumab is an investigational combination therapy being evaluated for the treatment of advanced solid tumors. YL201 is a B7-H3-targeted antibody-drug conjugate (ADC) developed by MediLink Therapeutics using its proprietary TMALIN® (Tumor Microenvironment Activable LINker-payload) technology. It consists of a human anti-B7-H3 monoclonal antibody conjugated to a camptothecin-based topoisomerase I inhibitor payload (YL0010014) with a drug-antibody ratio (DAR) of 8. Atezolizumab is a humanized monoclonal antibody that acts as a checkpoint inhibitor by binding to programmed death-ligand 1 (PD-L1), thereby preventing its interaction with PD-1 and B7.1 receptors. The combination is designed to provide synergistic anti-tumor activity by combining direct cytotoxic cell killing via the ADC with the restoration of T-cell mediated immune responses via PD-L1 blockade. It is currently in Phase 1 development for various solid tumors, including small cell lung cancer and nasopharyngeal carcinoma.
