Drug intelligence / Profile preview
YS2302018 + AZD0780 is an investigational oral combination therapy being developed by AstraZeneca for the treatment of dyslipidemia, hypercholesterolemia, and other cardiovascular diseases. The combination consists of two oral small-molecule agents: YS2302018, a preclinical lipoprotein(a) [Lp(a)] disruptor originally discovered by CSPC Pharmaceutical Group and licensed by AstraZeneca in 2024, and AZD0780 (laroprovstat), AstraZeneca's clinical-stage oral proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor. YS2302018 binds to apolipoprotein(a) [Apo(a)] to prevent its assembly with apolipoprotein B-100 [ApoB-100], thereby blocking the formation of atherogenic Lp(a) particles. AZD0780 binds to the C-terminal domain of PCSK9, preventing PCSK9-mediated degradation of low-density lipoprotein receptors (LDLR) and enhancing the clearance of LDL cholesterol. By combining these two complementary mechanisms, the therapy aims to simultaneously lower both LDL-C and Lp(a) levels, addressing key residual cardiovascular risk factors in patients with dyslipidemia.
