Overview

Zalifrelimab (AGEN1884) is a fully human monoclonal antibody that targets cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), acting as an immune checkpoint inhibitor to enhance the immune system's ability to recognize and destroy cancer cells. Pembrolizumab is a humanized monoclonal antibody targeting programmed cell death protein 1 (PD-1), also functioning as an immune checkpoint inhibitor by blocking the PD-1 pathway and preventing cancer cells from evading immune detection. The combination of zalifrelimab and pembrolizumab is being investigated in clinical trials for advanced solid tumors and metastatic non-small cell lung cancer (NSCLC). This combination aims to provide synergistic activation of the immune response against tumors by inhibiting two distinct checkpoints—CTLA-4 and PD-1[3][5][6].
