A combination regimen consisting of **zanidatamab**, a bispecific humanized monoclonal antibody targeting HER2 (human epidermal growth factor receptor 2), with **capecitabine** (an oral prodrug of 5-fluorouracil, a pyrimidine analog antimetabolite) and **oxaliplatin** (a platinum-based alkylating agent). Zanidatamab binds two distinct extracellular domains on HER2, inducing HER2 internalization, clustering, and multiple immune-mediated antitumor mechanisms such as antibody-dependent cellular cytotoxicity, complement-dependent cytotoxicity, and phagocytosis. This regimen is under investigation as a first-line treatment for patients with HER2-positive advanced or metastatic gastroesophageal adenocarcinoma and other HER2-expressing solid tumors. Capecitabine and oxaliplatin provide standard chemotherapy backbones for gastrointestinal cancers[1][2][3][4][5][6][7].
