Zanidatamab + paclitaxel is a combination therapy comprising the bispecific humanized monoclonal antibody zanidatamab and the cytotoxic chemotherapeutic paclitaxel. Zanidatamab targets and binds to two distinct extracellular epitopes of the HER2 receptor, leading to receptor crosslinking, blockade of HER2 signaling, and immune-mediated antitumor effects through antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP)[1][5]. Paclitaxel is a microtubule-stabilizing agent that inhibits mitosis. This combination is being evaluated for the treatment of HER2-expressing cancers, notably metastatic or unresectable gastroesophageal adenocarcinoma, biliary tract cancer, and HER2-positive breast cancer, primarily in settings where existing HER2-targeted therapies are limited or post-progression[2][3][5][6]. Zanidatamab's developer is Jazz Pharmaceuticals, and clinical studies have involved zanidatamab in combination with paclitaxel for efficacy and safety. The combination is investigational and not yet approved for any indication as of September 2025.
