Zanubrutinib + lenalidomide is an investigational combination therapy consisting of two small molecule drugs, zanubrutinib and lenalidomide. Zanubrutinib is a next-generation, highly selective Bruton’s tyrosine kinase (BTK) inhibitor that blocks B-cell receptor signaling, leading to inhibition of malignant B-cell proliferation and survival. Lenalidomide is an immunomodulatory agent with direct anti-tumor activity and indirect effects through enhancement of immune responses. The combination has been studied primarily in relapsed/refractory diffuse large B-cell lymphoma (DLBCL) and other lymphoid malignancies, showing promising efficacy and manageable safety profiles in early-phase clinical trials[4][7]. The regimen may be particularly relevant for patients who are unfit for standard chemotherapy or stem cell transplantation.
