Zasocitinib (TAK-279) is a highly selective, oral, allosteric inhibitor of tyrosine kinase 2 (TYK2), a member of the Janus kinase (JAK) family that mediates signaling for IL-23, IL-12, and Type I interferons. Developed by Takeda (following the acquisition of Nimbus Lakshmi), it is being investigated for various autoimmune conditions including plaque psoriasis and psoriatic arthritis. Repaglinide is a meglitinide-class insulin secretagogue used to improve glycemic control in adults with type 2 diabetes mellitus by closing ATP-dependent potassium channels on pancreatic beta cells. The combination of these two agents is specifically utilized in clinical pharmacology studies (such as NCT05826210) to assess the potential for zasocitinib to act as an inhibitor of CYP2C8 or OATP1B1, for which repaglinide serves as a sensitive probe substrate.
