zastaprazan + clopidogrel

JP-1366 + clopidogrel is a drug combination evaluated in clinical drug-drug interaction (DDI) studies, consisting of zastaprazan (JP-1366) and the antiplatelet agent clopidogrel. Zastaprazan is a novel potassium-competitive acid blocker (P-CAB) developed by Onconic Therapeutics (a subsidiary of Jeil Pharmaceutical). It suppresses gastric acid secretion by competitively and reversibly inhibiting the H+/K+-ATPase (proton pump) in gastric parietal cells, offering advantages over traditional proton pump inhibitors such as rapid onset and lack of food-effect or CYP2C19-related variability. Clopidogrel is a thienopyridine prodrug that, once metabolically activated, irreversibly inhibits the P2Y12 receptor on platelets to prevent aggregation. The co-administration is studied to ensure that the acid-blocking effects of zastaprazan do not clinically alter the pharmacokinetic profile or antiplatelet efficacy of clopidogrel, which is often used in patients who also require acid suppression.
