Overview

DW4421 is an investigational fixed-dose combination therapy developed by Daewon Pharmaceutical, comprising the potassium-competitive acid blocker (P-CAB) zastaprazan and the antacid sodium bicarbonate. Zastaprazan inhibits the gastric H+/K+-ATPase (proton pump) in a potassium-competitive and reversible manner, providing rapid and sustained suppression of gastric acid secretion. The inclusion of sodium bicarbonate is designed to neutralize gastric acid immediately upon administration, potentially accelerating the onset of the P-CAB's therapeutic effect and protecting it from acid-induced degradation in the stomach. The combination is currently in early-stage clinical development for the treatment of acid-related gastrointestinal disorders, including gastroesophageal reflux disease (GERD) and gastric ulcers.
