**Zastumotide + AS02B** is an investigational therapeutic cancer vaccine formulation containing recombinant MAGE-A3 antigen protein and the AS02B immunological adjuvant system. MAGE-A3 is a cancer-testis antigen expressed by a subset of melanomas, enabling antigen-directed immune recognition of tumor cells. AS02B enhances vaccine-induced cellular and humoral immunity to the recombinant antigen. The formulation was evaluated by the European Organisation for Research and Treatment of Cancer in the randomized open-label phase II EORTC 16032-18031 study in patients with MAGE-A3-positive unresectable stage III or stage IV M1a cutaneous melanoma. Clinical and immunologic activity was inferior to the corresponding MAGE-A3 plus AS15 regimen, and AS15 was selected for later development.
