Drug intelligence / Profile preview
zecdarasib + navlimetostat (INCB161734 + BMS-986504) is an investigational combination therapy being developed for the treatment of advanced or metastatic pancreatic ductal adenocarcinoma (PDAC). The combination consists of zecdarasib, an oral, selective, non-covalent small-molecule KRAS G12D inhibitor developed by Incyte, and navlimetostat, an oral, MTA-cooperative PRMT5 inhibitor developed by Bristol Myers Squibb (originally Mirati Therapeutics). Zecdarasib targets both the active (ON) and inactive (OFF) states of mutant KRAS G12D, which is a major oncogenic driver in pancreatic and colorectal cancers. Navlimetostat selectively targets the PRMT5-MTA complex, exploiting synthetic lethality in MTAP-deleted tumor cells. The combination leverages the synergistic potential of co-targeting the KRAS pathway and PRMT5/MTA axis in MTAP-deleted, KRAS-mutant pancreatic cancers. In late 2026, Incyte announced a clinical trial collaboration with Bristol Myers Squibb to evaluate this combination in a Phase 1b trial for patients with advanced or metastatic PDAC.
