Drug intelligence / Profile preview
Zedoresertib + temozolomide is an investigational combination therapy consisting of zedoresertib (Debio 0123), an oral, brain-penetrant, highly selective small-molecule inhibitor of WEE1 kinase, and temozolomide, an oral alkylating chemotherapy agent. Developed by Debiopharm (in-licensed from Almac Discovery), zedoresertib targets WEE1, a key regulator of the G2/M and S-phase DNA-damage checkpoints, preventing cancer cells from pausing to repair DNA damage and thereby sensitizing them to DNA-damaging treatments. Temozolomide is a standard-of-care chemotherapy that induces DNA damage. The combination is being evaluated in clinical trials (such as the Phase 1/2 study Debio 0123-GBM-105) for the treatment of glioblastoma (GBM), including recurrent or progressive GBM and newly diagnosed GBM (in combination with radiotherapy).
